=== Purification of a tagged protein === Another way to tag proteins is to engineer an antigen peptide tag onto the protein, and then purify the protein on a column or by incubating with a loose resin that is coated with an immobilized antibody. This particular procedure is known as immunoprecipitation. Immunoprecipitation is capable of generating an extremely specific interaction which usually results in binding only the desired protein. The purified tagged proteins can then easily be separated from the other proteins in solution and later eluted back into clean solution. When the tags are not needed anymore, they can be cleaved off by a protease. This often involves engineering a protease cleavage site between the tag and the protein. The main component of the tag is an intein, which cleaves off simply after a pH change. Tagless and pure target protein is then released into the elution buffer. == Structure == Cell-bound C3 and C5 convertase differ in their C3b requirement. C3-convertase (C3bBb) need only one molecule of C3b to form, whereas two or more C3b are required for generation of C5 convertase (C3bBb). It means, when C3b is randomly distributed on the surface of a cell, only C3 convertase activity appears after addition of Factors B and D. However, when C3b is distributed in clusters, C3 and C5 convertase activity is generated upon addition of Factors B and D. The classical pathway C5 convertase is composed of the fragments of complement proteins, C4b, C2a produced by cleavage mediated by C1 complex, and C3b produced by cleavage mediated by the classical pathway C3 convertase (C4bC2a). Factor B then binds to C3b and is cleaved by a plasma serine protease Factor D. The C3bBb complex (= alternative pathway C3 convertase) remains attached to the cell-surface. This complex might interact with another C3b and thus form the alternative pathway C5 convertase.

=== Wound Healing === Recent attention has been drawn to the remarkable wound-healing capabilities and excellent biocompatibility of keratin derived from human hair. While recombinant keratin proteins produced via recombinant DNA technology offer higher purity compared to extracted keratin, their wound-healing properties have remained unclear. Two recombinant trichocyte keratins—human type I hair keratin 37 and human type II hair keratin 81—were expressed using a bacterial expression system and subsequently forming recombinant keratin nanoparticles (RKNPs) through ultrasonic dispersion. It has been revealed that RKNPs significantly boosted cell proliferation and migration in laboratory settings. Moreover, when applied to dermal wounds in vivo, RKNPs facilitated improved wound healing, leading to enhanced epithelialization, vascularization, collagen deposition, and remodeling. Importantly, tests for in vivo biocompatibility showed no signs of systemic toxicity. RKNPs have potential as a promising approach for advancing wound healing and suggests new avenues for developing keratin-based biomaterials.

Suppliers normally quote a percentage of total chromatographic peak area, most often from reversed-phase HPLC. That figure says nothing about what the remaining percentage contains, and it depends on the detection wavelength used. A mass spectrometry result is a separate and stronger check on identity.
The methionine residue at the start of the chain is vulnerable to oxidation, producing a sulfoxide variant. Amide bonds can also hydrolyze, though more slowly under neutral conditions. Cold storage, oxygen exclusion and minimization of freeze-thaw cycles slow both processes but do not stop them.
A purity number on a certificate does not establish that a powder is the same product as a registered nasal medicine. Counter-ion content, residual solvents and peptide-related impurities may differ between the two. Independent verification is the only way to narrow that gap.
Solid peptide is best kept frozen at about minus 20 degrees Celsius in a sealed container with desiccant. It should be allowed to reach room temperature before opening to prevent condensation. Repeated warming and cooling of the same vial is discouraged.
