Once dissolved, the peptide is considerably less stable than the solid. Aqueous solutions are usually prepared at neutral to slightly acidic pH, filtered, and divided into single-use aliquots before freezing. Repeated freeze-thaw cycles are a common cause of avoidable loss and are best prevented by never refreezing a thawed aliquot. Adsorption to plastic and glass surfaces can lower the measured concentration of dilute solutions, particularly below roughly 0.1 mg/mL. Buffer choice, salt content, and container material all influence how much peptide remains detectable after storage.
Identity and purity are established with standard peptide methods. Reversed-phase HPLC with ultraviolet detection near 214 nm resolves the parent peak from deletion and truncation byproducts, and reports typically quote a main-peak percentage. Mass spectrometry by electrospray or MALDI-TOF confirms the expected molecular mass, while amino acid analysis or peptide mapping can verify composition when the sequence itself is in question. A certificate of analysis that pairs a chromatogram with a mass spectrum is more informative than a purity figure alone. Counter-ion content and residual solvents are separate specifications and are frequently omitted.
Development took place during the 1980s at institutes of the Russian Academy of Sciences, where peptide fragments of ACTH were screened for cognitive and neuroprotective effects. The compound received regulatory approval in Russia as a nasal preparation, marketed for conditions such as ischemic stroke, transient ischemic attacks, and optic nerve disorders. Registration in other countries has not followed. Most published clinical reports originate from a small number of Russian research groups, and independent replication outside that setting remains limited. The regulatory status therefore differs sharply between Russia and the rest of the world.
Laboratory work points toward modulation of neurotrophic signaling, particularly expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal tissue. Studies also describe effects on monoamine turnover, inflammatory mediators, and oxidative markers. These observations come mainly from animal models and cultured cells, so the causal chain in humans is not firmly established. Whether the reported molecular changes translate into measurable clinical benefit is an open question. Reviews generally present the mechanism as plausible rather than demonstrated.

Published research covers ischemic stroke, traumatic brain injury, cognitive impairment, optic nerve conditions and attention-related measures. Much of the human evidence comes from small trials conducted in one country, which limits how far the results generalize. Animal models supply the larger share of the data, and effects seen in rodents do not transfer automatically to people. Reviews have noted that methodological reporting is often incomplete, making it difficult to pool results or compare treatment schedules across studies.
Pharmacokinetic accounts emphasize rapid breakdown. After intravenous dosing the intact peptide disappears from blood within minutes, and nasal delivery produces low but measurable concentrations. Metabolites rather than the parent molecule may account for part of the observed activity, although the relative contribution is unresolved. Dosing in the literature varies widely and no optimal schedule has been agreed. These gaps are regularly cited as a reason the findings have not produced broad clinical adoption beyond the original research setting.
Proposed mechanisms center on neurotrophic signaling rather than on classical melanocortin receptor activation. Rodent experiments have reported shifts in the expression of brain-derived neurotrophic factor and nerve growth factor after administration, together with changes in the associated receptor systems. Several authors argue that the peptide acts largely through its degradation products and their interaction with peptidergic pathways, but this remains a hypothesis rather than a settled finding. No single molecular target has been identified in a way that the field broadly accepts.
Later generations of the molecule include an N-acetylated form and an amidated form, both marketed online alongside the parent peptide. These variants differ in terminal chemistry and stability, and they are frequently discussed in the same breath even though they have not been compared in controlled trials. Supply outside formal healthcare systems comes largely from laboratories that synthesize peptides to order. Purity and identity of these materials vary widely, and no single body oversees the international trade.
Semax is a synthetic heptapeptide developed in the Soviet Union during the 1980s by researchers at the Institute of Molecular Genetics in Moscow. It was designed as a truncated analog of adrenocorticotropic hormone, retaining only the fragment spanning residues four through ten. Investigators sought a peptide that would preserve the cognitive effects associated with ACTH while eliminating the hormonal stimulation of the adrenal cortex. The compound entered clinical use in Russia during the following decade.
Russian regulatory authorities approved the peptide for nasal administration, and it remains listed in the national pharmacopoeia under several trade names. Documented indications include acute ischemic stroke, transient ischemic attacks, traumatic brain injury, and certain ophthalmological and neurological conditions. Physicians also prescribe it for cognitive complaints in older patients, although the evidence base for that use is thinner. Outside Russia and a few neighboring states, the substance is not an approved medicine and is sold instead as a research chemical.
Common degradation pathways include oxidation of the methionine side chain, hydrolysis of the peptide backbone, and aggregation under unfavourable pH or concentration. Stability studies typically monitor the main peak by chromatography and report total related substances as a percentage. Because no official monograph exists, acceptance criteria vary between laboratories, and reported purity values are not directly comparable across suppliers. Analysts therefore document the method, column, and detection wavelength alongside each result, and open questions remain about how much biological activity the oxidised forms retain.
Solid semax is typically supplied as a lyophilised powder that is hygroscopic and sensitive to moisture, light, and repeated temperature cycling. Long-term storage of the dry peptide is generally recommended at approximately -20 degrees Celsius, while shorter working periods may use refrigeration at 2 to 8 degrees Celsius. Vials should remain tightly closed and desiccated when brought to room temperature, because condensation can damage the material before it is weighed. Dividing a batch into aliquots is preferable to thawing one container repeatedly.
This process is run thousands or millions of times. The number of times a single tree is visited during the course of the chain is an approximation of its posterior probability. Some of the most common algorithms used in MCMC methods include the Metropolis–Hastings algorithms, the Metropolis-Coupling MCMC (MC³) and the LOCAL algorithm of Larget and Simon.

=== Mammals === Several groups of ungulates have independently reduced or lost side digits on their feet, often leaving one or two digits for walking. That name comes from their hooves, which have evolved from claws several times. For example, horses have one walking digit and domestic bovines two on each foot. Various other land vertebrates have also reduced or lost digits. Similarly, laurasiathere perissodactyls and afrothere paenungulates have several features in common, to the point of there being no obvious distinction among basal taxa of both groups. Many aquatic mammals or marine mammals independently came to have adaptations to live in water, such as similar-looking tail flukes in dugongs and whales. Unrelated herbivores and carnivores have adapted to marine and freshwater environments. The flipper forelimbs of marine mammals (cetaceans, pinnipeds, and sirenians) are a classic example of convergent evolution. There is widespread convergence at the gene level. Distinct substitutions in common genes created various aquatic adaptations, most of which also constitute parallel evolution because the substitutions in question are not unique to those animals. The pronghorn of North America, while not a true antelope and only distantly related to them, closely resembles the true antelopes of the Old World, both behaviorally and morphologically. It also fills a similar ecological niche and is found in the same biomes. Members of the two clades Australosphenida and Theria evolved tribosphenic molars independently.

== Mechanism of action == Melanotan II acts as a non-selective agonist of the melanocortin receptors MC1, MC3, MC4, and MC5. Melanotan II produces melanogenesis by activation of the MC1 receptor, whereas its clinically documented sexual effects are thought to be related to its ability to activate the MC4 receptor (though the MC3 is thought to also possibly be involved). Melanotan II is partly metabolised into Bremelanotide, a medication used to treat low sexual desire. Other effects of melanotan II, mostly regarded as adverse effects, include flushing, nausea, vomiting, stretching, yawning, and loss of appetite (the last via activation of MC4).

==== ssDNA vs. dsDNA ==== Most DNA molecules are actually two polymer strands, bound together in a helical fashion by noncovalent bonds; this double-stranded (dsDNA) structure is maintained largely by the intrastrand base stacking interactions, which are strongest for G,C stacks. The two strands can come apart—a process known as melting—to form two single-stranded DNA (ssDNA) molecules. Melting occurs at high temperatures, low salt and high pH (low pH also melts DNA, but since DNA is unstable due to acid depurination, low pH is rarely used). The stability of the dsDNA form depends not only on the GC-content (% G,C basepairs) but also on sequence (since stacking is sequence specific) and also length (longer molecules are more stable). The stability can be measured in various ways; a common way is the melting temperature (also called Tm value), which is the temperature at which 50% of the double-strand molecules are converted to single-strand molecules; melting temperature is dependent on ionic strength and the concentration of DNA. As a result, it is both the percentage of GC base pairs and the overall length of a DNA double helix that determines the strength of the association between the two strands of DNA. Long DNA helices with a high GC-content have more strongly interacting strands, while short helices with high AT content have more weakly interacting strands. In biology, parts of the DNA double helix that need to separate easily, such as the TATAAT Pribnow box in some promoters, tend to have a high AT content, making the strands easier to pull apart.
=== Industrial === Industrial biotechnology (known mainly in Europe as white biotechnology) is the application of biotechnology for industrial purposes, including industrial fermentation. It includes the practice of using cells such as microorganisms, or components of cells like enzymes, to generate industrially useful products in sectors such as chemicals, food and feed, detergents, paper and pulp, textiles and biofuels. In the current decades, significant progress has been done in creating genetically modified organisms (GMOs) that enhance the diversity of applications and economical viability of industrial biotechnology. By using renewable raw materials to produce a variety of chemicals and fuels, industrial biotechnology is actively advancing towards lowering greenhouse gas emissions and moving away from a petrochemical-based economy. Synthetic biology is considered one of the essential cornerstones in industrial biotechnology due to its financial and sustainable contribution to the manufacturing sector. Jointly biotechnology and synthetic biology play a crucial role in generating cost-effective products with nature-friendly features by using bio-based production instead of fossil-based. Synthetic biology can be used to engineer model microorganisms, such as Escherichia coli, by genome editing tools to enhance their ability to produce bio-based products, such as bioproduction of medicines and biofuels. For instance, E.

In particular, Jung argued that dreams are best used in analysis with the methods of amplification and active imagination, rather than by interpretation through free association (as Freud proposed). Jung saw dreams as serving individuation by making unconscious material available to the whole personality. Jung originated the idea of therapy as a dialectical process - a two-way process where both parties are equally involved. He saw it was important to see the patient as a real person rather than a sick inferior. In this way, he also saw himself as not having all the answers and saw the process of a patient finding their own answers as much more valuable. He made a point of not being dogmatic in his approach and treating each patient as the individual they are. As such, he disregarded group therapies. Jung saw analysis as having four stages:
Supercomputers – including Summit and Fugaku – have been used to explore potential treatments by running simulations with data on already-approved medications. Two early examples of supercomputer consortia are listed:
== Mechanical properties == As noted above, the (two-dimensional) density of graphene is 0.762 mg per square meter. Graphene is the strongest material ever tested, with an intrinsic tensile strength of 130 GPa (19,000,000 psi) (with representative engineering tensile strength ~50-60 GPa for stretching large-area freestanding graphene) and a Young's modulus (stiffness) close to 1 TPa (150,000,000 psi). The Nobel announcement illustrated this by saying that a 1 square meter graphene hammock would support a 4 kg cat but would weigh only as much as one of the cat's whiskers, at 0.77 mg (about 0.001% of the weight of 1 m2 of paper). Large-angle bending of graphene monolayers with minimal strain demonstrates its mechanical robustness. Even under extreme deformation, monolayer graphene maintains excellent carrier mobility. The spring constant of suspended graphene sheets has been measured using an atomic force microscope (AFM). Graphene sheets were suspended over SiO2 cavities where an AFM tip was used to apply stress to the sheet to test its mechanical properties. Its spring constant was in the range 1–5 N/m and the stiffness was 0.5 TPa, which differs from that of bulk graphite. These intrinsic properties could lead to applications such as NEMS as pressure sensors and resonators. Due to its large surface energy and out of plane ductility, flat graphene sheets are unstable with respect to scrolling, i.e. bending into a cylindrical shape, which is its lower-energy state.
=== Data breach === According to an October 2020 report, for months, the company stored hundreds of massive spreadsheets containing sensitive patient data in an Amazon Web Services storage bucket without a password. TechCrunch was the first news source to report the data breach.
== Triple dilution method == To avoid contamination of the mass spectrometer with the isotopically enriched spike, an additional blend of the primary standard (A*) and the spike (B) can be measured instead of measuring the enriched spike (B) directly. This approach was first put forward in the 1970s and developed in 2002.

Each freeze-thaw cycle can degrade a fraction of the peptide and promote aggregation. Dividing a stock solution into single-use volumes removes repeated cycling as a variable. It also limits the time a thawed solution spends at room temperature.
A high main-peak percentage says little about identity, counter-ion content, or residual solvents. It also does not distinguish an isomer or a closely eluting analog. Mass confirmation and the accompanying documentation cover those gaps.
Salt form changes mass, solubility, and hygroscopicity, and acetate and trifluoroacetate salts behave differently in solution. Storage guidance still centers on -20 °C for powder and lower temperatures for solutions. The certificate should state which salt is present.
Semax is a synthetic seven-amino-acid peptide derived from a fragment of corticotropin. It is used in Russia as a nasal preparation, while elsewhere it is studied as a research compound. It is not approved as a medicine in most countries.
