=== Exhaustion === T cell exhaustion is a poorly defined or ambiguous term. There are three approaches to its definition. "The first approach primarily defines as exhausted the cells that present the same cellular dysfunction (typically, the absence of an expected effector response). The second approach primarily defines as exhausted the cells that are produced by a given cause (typically, but not necessarily, chronic exposure to an antigen). Finally, the third approach primarily defines as exhausted the cells that present the same molecular markers (typically, programmed cell death protein 1 [PD-1])." Indeed, it is now starting to emerge that exhaustion might not be the only T cell dysfunctional state. In fact, tolerization, anergy, cell death, ignorance, senesence and exclusion have recently emerged as additional sources and/or states of T cell dysfunction in cancer and chronic viral infection. Dysfunctional T cells are characterized by progressive loss of function, changes in transcriptional profiles and sustained expression of inhibitory receptors. At first, cells lose their ability to produce IL-2 and TNFα, which is followed by the loss of high proliferative capacity and cytotoxic potential, and eventually leads to their deletion. Exhausted T cells typically indicate higher levels of CD43, CD69 and inhibitory receptors combined with lower expression of CD62L and CD127. ==== Distribution ==== Psilocin, the active form of psilocybin, is extensively distributed to all tissues through the bloodstream. Its volume of distribution is 505 to 1,267 L. Psilocybin itself is hydrophilic due to its phosphate group and cannot easily cross the blood–brain barrier. Conversely, psilocin is lipophilic and readily crosses the blood–brain barrier to exert effects in the central nervous system. == Ion-exchange chromatography == DEAE-C is a weak anion exchanger. This exchange is utilized to separate proteins that have faintly differing charges. Like all anion exchangers, the resin carries a positive charge that interacts favorably with negative charges. The positive charge of DEAE cellulose is due to a protonated amine group. To ensure that the resin is protonated and positively charged, the chromatography should be performed at least 2 pH units below the pKa of the amine group, 10. The strength of the bond between the resin and protein is highly dependent on the pH range in the column and the pI of the protein of interest. The resin is a weak exchanger because it is only partially ionized over most pH values, and an efficient separation with DEAE-C chromatography requires a specific, narrow pH range. Cellulose, dextran, agarose, and other insoluble complexes are unaffected because they compose inert matrices, hence why they are so often derivatized with strong and weak cation and anion exchangers in chromatography.
